Identification of Human Chromosome Segments that Have High Homology with Rat Genomic DNA
نویسندگان
چکیده
This study was conducted to determine the location of DNA segment with homology to the rat conserved genomic DNA in human chromosomes. The labeled rat genomic DNA was hybridized with normal human (male) metaphases. The study of 74 metaphases after fluorescence in situ hybridization showed 371 twin-spot signals on human chromosomes. Statistical analysis indicated that the specific accumulation of signals on 1q22-qter, 2p2, 3p21-p23, 4q3, 6q2, 8p12-pter, 11p12-pter, 11q12-qter, 12q2, 13p, 15p, 16q2, 21q12-qter, Yq1-qter, and Xq2 was not random. Results of stepwise multiple linear regressions indicated that number of mapped oncogenes (Beta = 1.092; t = 7.552; P<0.001) and density of mapped oncogenes on chromosomes (Beta = -0.832; t = -5.751; P<0.001) have significant effects on number of double-spots on human chromosomes. These data reflects the evolutionary conservation between rat DNA and human DNA at the above-mentioned bands. Iran. Biomed. J. 9 (1):

متن کاملIdentification of High Crash Road Segment using Genetic Algorithm and Dynamic Segmentation
This paper presents an evolutionary algorithm for recognizing high and low crash road segments using Genetic Algorithm as a dynamic segmentation method. Social and economic costs as well as physical and mental injuries make the governments perceiving to road safety indexes in order to diminish the consequences of road accidents. Due to the limitation of budget for safety...
متن کاملCloning and sequencing of Toxoplasma gondii major surface antigen (SAG1) gene
Genetic typing methods of T. gondii strains have been extensively perfected in recent years. From a technical point of view, many tools usable for genetic studied on single-copy loci have been used: RFLP, PCR-RFLP, sequencing, RAPD-PCR and isoenzyme analysis. We described the cloning and sequence analysis of the gene which encodes the major surface antigen (SAG1 or P30) of T. gondii. SAG1 is ...
متن کاملMolecular hybridization under conditions of high stringency permits cloned DNA segments containing reiterated DNA sequences to be assigned to specific chromosomal locations.
Identifying the specific DNA sequences involved in the chromosomal abnormalities in developmental and neoplastic diseases may be essential to understanding the molecular biology of these disorders. The use of recombinant DNA techniques in conjunction with rodent-human hybrid cells makes it possible to assign chromosomal locations to specific DNA sequences. However, the ubiquitous presence of re...
متن کاملDevelopment of an Alu-PCR Amplified YAC Probe Suitable for Enumeration of Chromosome 13 on Uncultured Lymphocytes and Amniocytes by Fluorescence in situ Hybridization
The main objective of the present study was to develop an efficient and reliable probe to be routinely used for detection of chromosome 13 copy numbers by interphase FISH. To achieve this, a Yeast Artificial Chromosome (YAC) containing sequences specific for human 13q12 (744D11), was cultured and the whole yeast genomic DNA was extracted. The human insert within the isolated DNA was amplified b...
